RESUMEN

Accurate and efficient prediction of drug-target interaction (DTI) is critical to advance drug development and reduce the cost of drug discovery. Recently, the employment of deep learning methods has enhanced DTI prediction precision and efficacy, but it still encounters several challenges. The first challenge lies in the efficient learning of drug and protein feature representations alongside their interaction features to enhance DTI prediction. Another important challenge is to improve the generalization capability of the DTI model within real-world scenarios. To address these challenges, we propose CAT-DTI, a model based on cross-attention and Transformer, possessing domain adaptation capability. CAT-DTI effectively captures the drug-target interactions while adapting to out-of-distribution data. Specifically, we use a convolution neural network combined with a Transformer to encode the distance relationship between amino acids within protein sequences and employ a cross-attention module to capture the drug-target interaction features. Generalization to new DTI prediction scenarios is achieved by leveraging a conditional domain adversarial network, aligning DTI representations under diverse distributions. Experimental results within in-domain and cross-domain scenarios demonstrate that CAT-DTI model overall improves DTI prediction performance compared with previous methods.

AIM: To explore the employment intention and career planning of male nursing students at different levels of colleges and universities and provide references for formulating individualized training content. DESIGN: Phenomenological research method in qualitative research. METHODS: Using a phenomenological research method, 15 male nursing students from three levels of colleges and universities were interviewed in a one-to-half structure, and the data were analysed using NVivo12.0 software and the Colaizzi seven-step analysis method. RESULTS: Four themes and 10 sub-themes were extracted. The four themes included professional identity experience, nursing learning experience, career expectation and planning and educational needs. The 10 sub-themes included the negative effects of traditional impressions, the gradual establishment of professional identity, public health events promote professional identity, negative experiences in learning, positive growth from learning, varied career plans, career expectations and influencing factors, a reasonable schedule, diversity of content requirements and the positive effect of teaching teachers. CONCLUSIONS: Influenced by many factors, the employment intention and career planning of male nursing students at different higher education levels are varied. Schools and clinical practice units should strengthen the training of employment intention and career planning and make reasonable time adjustments to the training content. PUBLIC CONTRIBUTION: In total, 15 male interns accepted our interview request. We utilized their break time to conduct the interviews, resulting in significant contributions to the content of the article.

RESUMEN

Sepsis is a life-threatening organ dysfunction syndrome caused by host response disorders due to infection or infectious factors and is a common complication of patients with clinical trauma, burns, and infection. Resveratrol is a natural polyphenol compound that is a SIRT-1 activator with anti-inflammatory, antiviral, antibacterial, antifungal inhibitory abilities as well as cardiovascular and anti-tumor protective effects. In recent years, some scholars have applied resveratrol in animal models of sepsis and found that it has an organ protective effect and can improve the survival time and reduce the mortality of animals with sepsis. In this study, Medline (Pubmed), embase, and other databases were searched to retrieve literature published in 2021 using the keywords "resveratrol" and "sepsis," and then the potential of resveratrol for the treatment of sepsis was reviewed and prospected to provide some basis for future clinical research.

RESUMEN

Hydrazide-modified graphene quantum dots (HM-GQDs) obtained by refluxing GQDs with hydrazine hydrate were hybridized with gold nanoparticles (AuNPs) through a redox reaction between HM-GQDs and AuCl4(-). The obtained nano-hybrids (HM-GQD-AuNPs) possess the unique electrochemiluminescence (ECL) properties of HM-GQDs and the easy self-assembly with some bio-molecules of AuNPs, which have great potential applications in bio-sensors. HM-GQD-AuNPs were modified on a glassy carbon electrode to develop a novel ECL immunosensor of carcinoembryonic antigen (CEA) as a model target analyte. Due to the increment of electron-transfer resistance after immunoreaction, the ECL intensity decreased as the concentration of CEA was increased. The linear response range was between 0.02 and 80 ng mL(-1), and the detection limit was 0.01 ng mL(-1).

Proteases and nucleases are enzymes heavily involved in many important biological processes, such as cancer initiation, progression, and metastasis; hence, they are indicative of potential diagnostic biomarkers. Here, we demonstrate a new label free and sensitive electrochemiluminescent (ECL) sensing strategy for protease and nuclease assays that utilize target-triggered desorption of programmable polyelectrolyte films assembled on graphite-like carbon nitride (g-C3N4) film to regulate the diffusion flux of a coreactant. Furthermore, we have built Boolean logic gates OR and AND into the polyelectrolyte films, capable of simultaneously sensing proteases and nucleases in a complicated system by breaking it into simple functions. The developed intelligent permeability controlled enzyme sensor may prove valuable in future medical diagnostics.

Two-dimensional graphite-like carbon nitride nanosheets (g-C3N4 NSs) were hybridized with gold nanoparticles (Au NPs) to construct an electrochemiluminescence (ECL) immunosensor. The prepared Au NP-functionalized g-C3N4 NS nanohybrids (Au-g-C3N4 NHs) exhibit strong and stable cathodic ECL activity compared to g-C3N4 NSs due to the important roles of Au NPs in trapping and storing the electrons from the conduction band of g-C3N4 NSs, as well as preventing high energy electron-induced passivation of g-C3N4 NSs. On the basis of the improved ECL stability and ECL peak intensity of the Au-g-C3N4 NHs, a novel ECL immunosensor was developed to detect carcinoembryonic antigen (CEA) as a model target analyte. The ECL immunosensor has a sensitive response to CEA in a linear range of 0.02-80 ng mL(-1) with a detection limit of 6.8 pg mL(-1). Additionally, the proposed immunosensor shows high specificity, good reproducibility, and long-term stability.

OBJECTIVE: To observe the effect of Rupifang Extract in external use on expression of proto-oncogenes her2 and tumor suppression genes p16 in rat breast tissues of mammary hyperplasia model. To explore the mechanisms of Rupifang Extract in external use for preventing and treating mammary hyperplasia. METHODS: Thirty virginal female Wistar rats were randomized into 5 groups, 6 in each, A: blank control group; B: model group; C: the low dose group of Rupifang; D: the middle dose group of Rupifang; and E: The high dose group of Rupifang. The mammary hyperplasia rat models were produced by injecting estradiol benzoate and progesterone and irritating by tail nipping. Drug intervention was also launched during the model formation. After 30 days, the expression of her2 and p16 in breast tissues of rats in each group were detected by the SP immunohistochemical method. RESULTS: Compared with Blank control group, the expression of her2 in breast tissues in Model group was higher, and the expression of p16 was lower (P < 0.05 or P < 0.01). After intervention with Rupifang Extract, compared with Model group, the expression of her2 in breast tissues in Rupifang groups was lower, and the expression of p16 higher (P < 0.05 or P < 0.01). CONCLUSION: The mechanisms of Rupifang Extract in external application for preventing and treating mammary hyperplasia may be reducing the expression of proto-oncogenes her2 and increasing the expression of tumor suppression genes p16.
